Indications/Uses
Peripheral & central neuropathic pain & generalized anxiety disorder (GAD) in adults. Adjunctive therapy in adults w/ partial seizures w/ or w/o secondary generalization. Adjunctive therapy for adults w/ partial onset seizures.
Dosage/Direction for Use
150-600 mg daily in 2 or 3 divided doses. Neuropathic pain Initially 150 mg daily given as 2 or 3 divided doses, may be increased to 300 mg daily after an interval of 3-7 days. Max: 600 mg daily after an additional 7-day interval. Epilepsy Initially 150 mg daily given as 2 or 3 divided doses, may be increased to 300 mg daily after 1 wk. Max: 600 mg daily after an additional wk. GAD Initially 150 mg daily, may be increased to 300 mg daily after 1 wk. Following an additional wk, may be increased to 450 mg daily. Max: 600 mg daily after an additional wk. Renal impairment CrCl ≥60 mL/min 150 mg bid or tid. Max: 600 mg daily; ≥30-<60 mL/min 75 mg bid or tid. Max: 300 mg daily; ≥15-<30 mL/min 25-50 mg once daily or bid. Max: 150 mg daily; <15 mL/min 25 mg once daily. Max: 75 mg daily. Supplementary dose following haemodialysis 25 mg as a single dose. Max: 300 mg daily.
Administration
May be taken with or without food.
Special Precautions
Discontinue immediately if angioedema & hypersensitivity occurs. Patients w/ previous episode of angioedema & those taking other drugs associated w/ angioedema (eg, ACE inhibitors). W/draw gradually over a min of 1 wk. Peripheral edema. Co-administration w/ thiazolidinediones. May impair ability to perform tasks eg, driving or operating machinery. Childn <12 yr & adolescents (12-17 yr). Elderly ≥65 yr.
Adverse Reactions
Dizziness & somnolence. Increased appetite; euphoric mood, confusion, irritability, decreased libido, disorientation, insomnia; ataxia, abnormal coordination, tremor, dysarthria, memory impairment, disturbance in attention, paraesthesia, sedation, balance disorder, lethargy, headache; blurred vision, diplopia; vertigo; vomiting, dry mouth, constipation, flatulence; erectile dysfunction; abnormal gait, feeling drunk, fatigue, peripheral edema, edema; increased wt.
Drug Interactions
May increase sedative effects w/ alcohol & drugs that cause sedation. May increase occurrence of wt gain, fluid retention & possibly heart failure w/ pioglitazone & rosiglitazone.
